What Is UV-Vis Spectrophotometer?

About Ultraviolet

When a white light is projected into a prism or a slit, it will be dispersed into spectrum. Lower frequency red light with less energy, at one end of the spectrum, while the higher frequency purple light with richer energy, at the other end. And out of this side, it is the region of the invisible electromagnetic radiation. In addition to ultraviolet (UV), there are other forms of high energy invisible lights. The UV radiation can hurt your skin, when you stand in the sun light.

According to the definition in ISO 21348:2007 by International Organization for Standardization (ISO), we can split the ultraviolet into 4 types (segments), according to the wavelength (the energy), they are NUV, MUV, FUV, and EUV. Extreme ultraviolet (EUV), opposites to near ultraviolet, at the end of UV region, where is close X-rays, and it is the most energetic one among the 4 types. And middle ultraviolet (MUV) and far ultraviolet (FUV) are in between the EUV and NUV. According to ISO 21348:2007, the segments and their wavelengths are defined in the below table:

Extreme Ultraviolet EUV 10 nm - 121 nm
Far Ultraviolet FUV 122 nm - 200 nm
Middle Ultraviolet MUV 200 nm - 300 nm
Near Ultraviolet NUV 300 nm - 400 nm

Note: The wavelength of visible light region is 380 nm – 760 nm.

The Function of UV-Vis Spectroscopy

UV / Vis spectrophotometer uses visible light and ultraviolet to analyze the chemical structure of substance. A spectrophotometer is a special type of spectrometer, which is used to measure the intensity of light, and the intensity is proportional to the wavelength. When ultraviolet project to various organic compounds, these compounds will absorb it. So, you can use UV / Vis spectrophotometer to measure the absorption of a compound by the result, and have its molecular structure, as well as the related information.

The Types of UV-Vis Spectrophotometer

UV / Vis spectrophotometer has two types, single-beam and dual-beam. The single-beam UV-Vis spectrophotometer has only one cuvette, therefore, the Treatment Group and the Control Group samples can one be measured one after the other. With the double-beam UV-Vis spectrophotometer, you can measure the both groups simultaneously. So that the double beam UV-Vis spectrophotometer is more accurate, because you don’t need to recalibrate the instrument, before you measure the second sample.
